The intern will use aPKC inhibitor as pharmacologic tools on the established assays to validate aPKC's role in regulating the PAR3/Numb à p120catenin/E-cadherin pathway. If successfully executed, the intern's project may provide new pharmaco-dynamic reporter assays of aPKC function relevant to its role in tumor growth and maintenance and may provide a framework for establishing more clinically translatable and relevant biomarker assays.
Project Overview:
Atypical protein kinase C enzymes function as part of the PAR polarity complex composed of PAR3 and PAR6. The PAR complex participates in cellular polarity events required for normal cell migration and epithelial cell apical-basal polarity. A growing body of evidence suggests that dysregulation of cell polarity may promote epithelial-mesenchymal transition events linked to tumor initiation and metastasis. Recent in vitro biochemical and cell biology experiments employing genetic knock-down have shown a link between aPKC and the proposed tumor suppressor Numb, a membrane-bound substrate of aPKC involved in regulating internalization and trafficking of adhesion receptors (eg integrins and E-cadherin). To date, no studies have employed selective potent aPKC inhibitors to pharmacologically validate its role in Numb regulation. MRL-Boston Oncology has developed selective aPKC small molecule inhibitors well suited for in vitro pharmacology experiments. The intern's project goal will be to pharmacologically validate aPKC's role in regulating Numb-mediated events. Building on literature precedents and employing new assay approaches (eg DuoLink PLA) the intern will help to establish assays that measure aPKC/Par6 à PAR3/Numbà p120catenin/E-cadherin trafficking and protein interactions in relevant tumor cell lines.
This is a 9-11 week, paid internship beginning in June 2012. Housing will be provided to those who meet distance requirement.

The Department of Molecular Biomarkers is seeking a highly motivated individual to join our Biological Mass Spectrometry group. This newly created position requires execution on quantitative proteomic strategies to measure changes in protein expression, and posttranslational modifications in response to stimuli to support the oncology and immunology franchises. Core expertise in mass spectrometry and analytical instrumentations are required as are excellent analysis skills. Preference will be given to those applicants with experience in high resolution mass spectrometry and the use of stable isotopes as internal standards. A Ph.D. in Chemistry/Protein Chemistry or Master's degree with 3 years of experience is required. Understanding of flow cytometry fundamentals and antibody conjugation is a plus. It is expected that the successful candidate will publish independently in first tier peer reviewed journals.

The Informatics and Analysis (I&A) department within Discovery and Pre-Clinical Sciences supports the application of genomic and genetic approaches to biomarker and target discovery to all of MRL therapeutic area franchises. The Research Solutions and Bioinformatics team within I&A works closely with drug program biologists and clinicians to design and analyze data from large scale molecular profiling studies, with an emphasis on biological pathway analysis and interpretation.
Position Description: Highly motivated bioinformatics scientist with a strong background in immunology or oncology. The successful candidate will collaborate closely with pre-clinical and clinical drug programs teams as well as data analysts. This role is at the crucial interface of data analysis and biology and the candidate will be required to master both ends.
The responsibilities of the candidate include but are not limited to:
* Analyzing and integrating large-scale experimental data sets including gene expression, next-generation sequencing and RNAi screens to identify biomarkers of drug response, drug mechanism of action and drug resistance mechanisms. * Performing biological pathway and network analysis to form hypothesis and collaborating with biologists to suggest further experiments and validate key findings. * Designing, creating and maintaining databases to host, analyze and share raw and analysis data. * Devise biomarker strategies that leverage profiling technologies. * Author sections of biomarker development plans and clinical protocols.

As the Safety Assessment (SA) representative to Merck Research Laboratories discovery research teams at the Boston, MA site, the SA discovery scientist leads the collaboration to define the preclinical SA strategy for programs prior to the approval of First-In-Man enabling GLP toxicity studies. With excellent leadership and collaborative skills, a broad expertise in preclinical safety assessment and an intimate understanding of the science and objectives of the assigned discovery programs, the SA discovery scientist assesses the safety risk using available literature, internal and external data and guides discovery and SA scientists to execute on the appropriate non-GLP SA studies to support selection of therapeutic targets and new molecular entities (NCE) with the highest probability of success for the intended indication. In addition, the SA discovery scientist is part of the scientific team that discovers and implements novel biomarkers and screening strategies. The SA discovery scientist is encouraged to participate in SA functional group activities aligned with their area of scientific expertise.
* Uses literature and data-base resources to understand and define safety risks associated with therapeutic targets. * Defines the appropriate in silico approaches and in vitro and in vivo studies, including design, dose selection, compound requirements and timing to help innovate and select NCE with the highest probability of success. * Collaborates with SA and Discovery scientists to assure the appropriate conduct of in silico assessments and in vitro and in vivo studies that support discovery programs. * Effectively communicates the safety de-risking strategies, study data and conclusion to discovery teams, governance committees and management. * Collaborates with the discovery and SA scientists to address all scientific issues including assessment of efficacy, metabolism, formulation and purity of drug product and substance. * Provides consultation and guidance to discovery scientists on complex scientific problems and in general is available for consultation and advice on research approaches. * Works closely with the SA development compound and therapeutic leaders to assure smooth transition and knowledge transfer for NCE approved for the development and understands safety data from NCE in development to build appropriate derisking strategies to support the innovation of improved back-up molecules. * Provides due diligence support for licensing efforts of targets and NCE in the discovery space. * May contribute as study pathologist to exploratory and mechanistic studies supporting discovery teams, if compatible with training and experience.
Qualifications
Education:
* MD, DVM and/or PhD. Degree from an accredited college or university with graduate and/or post-graduate work and/or residency training in Toxicology, Pathology, or related field.
Required:
* 10 or more years experience in Toxicology or related field and 5 or more years of demonstrated experience as pre-clinical SA scientist and team representative in the pharmaceutical industry with an extensive expertise and/or an excellent understanding of regulatory expectations, genetic toxicology, safety pharmacology, reproductive and developmental toxicology and general toxicology and pathology.
